Measuring, Shortening and Functionalizing Carbon Nanotube Tipped AFM Probes for DNA Sequencing

چکیده
A method is presented to indirectly measure and shorten carbon nantoubes grown from the tips of atomic force microscope (AFM) probes. The measurement component exploits the nanotubes ability to elastically buckle and requires only those signals and actuators available on a standard AFM. The shortening operation is facilitated by electric arcing using a conducting niobium substrate. The shortening operation produces stable carbon nanotubes at the tips of AFM probes which can then be functionalized for use as high-resolution, chemically-specific force probes. An application to AFM-based DNA sequencing is also discussed.

A single molecule of DNA is denatured and immobilized on an atomically flat substrate, and a carbon nanotube force probe functionalized with a nucleotide is scanned along the molecule to detect locations of the probe nucleotide’s complement. Discussion is offered regarding the preparation of single-walled carbon nanotube AFM probes and their chemical attachment of DNA nucleotides. Additionally,...

Atomic force microscopy (AFM) has great potential as a tool for structural biology, a field in which there is increasing demand to characterize larger and more complex biomolecular systems. However, the poorly characterized silicon and silicon nitride probe tips currently employed in AFM limit its biological applications. Carbon nanotubes represent ideal AFM tip materials due to their small dia...

The invention of atomic force microscopy (AFM) is having a great impact on various areas, such as nano metrology, materials science, surface science and biology (Binnig et al., 1986). The lateral resolution of AFM is mainly determined by the probe’s shape and physical property, especially the geometry and dimension of the probe end. Conventional AFM probe is pyramidal shape by micro-fabrication...

Magnetic force microscope ( MFM ) is a powerful technique for mapping the magnetic force gradient above the sample surface. Herein, single-wall carbon nanotubes (SWCNT) were used to fabricate MFM probe by dielectrophoresis method which is a reproducible and cost-effective technique. The effect of induced voltage on the deposition manner of carbon nanotubes (CNT) on the atomic force microscope (...
